I think it is really easy to make

The colors are invisible circle sprites.
When you overlap a invisible circles with the mouse the circles will made visible.

hmm it might be it, but i must had to put a lot of these objects invisible and the example you can color with very high pixel-precise

wow! your example looks more precise than in my link:) To fill the star you have to create a lot of blue circular objects. Only if in the case of larger surface coloring, the coloring will not display a less smoothly?
